    Abstract: 本研究的目的為發展一個壓電免疫感測器並應用於結核病體液免疫檢驗。選用結核菌特有的專一性蛋白CFP-10為辨識分子,以區別因為接種過卡介苗所造成的偽陽性,提升檢驗方法的準確性。試驗分為兩部分,第一部分為免疫感測器製備方法的確立。分別以長碳鏈(16碳)硫醇及短碳鏈(2碳)硫醇修飾壓電晶片金電極表面,試驗結果顯示長碳鏈硫醇所修飾的表面,降低表面的立體障礙,有較佳的檢測靈敏度。第二部分探討應用免疫感測器於兔子血清抗體檢測的可行性。結果顯示在適當稀釋倍數範圍,兔子血清抗體濃度在不同稀釋倍數下皆有一致性。因此,預期本研究提出的方法,能為現有結核病體液免疫檢測方法,提供另一個選擇。
    The purpose of this study is to develop a piezoelectric immunosensor and apply it to the detection of tuberculosis humoral immunity. The specific protein CFP-10 unique to Mycobacterium tuberculosis was selected as the identification molecule to distinguish false positives caused by BCG vaccination and to improve the accuracy of the test method. The experiment was divided into two parts, the first part was the establishment of the immunosensor preparation method. The long carbon chain (16 carbon) thiol and short carbon chain (2 carbon) thiol were used to modify the surface of the gold electrode of the piezoelectric chip. The test results showed that the surface modified by the long carbon chain thiol had better detection sensitivity. The second part discussed the feasibility of using the immunosensor to detect antibodies in rabbit serum. The results showed that in the appropriate dilution range, the rabbit serum antibody concentrations were consistent under different dilution ratios. Therefore, it is expected that the method proposed in this study can provide another option for the existing TB humoral immunoassay.
