本研究係以幾丁聚醣與N-環氧基2,3丙基-N-甲基-N,N-二烯丙基甲基磺酸化銨 (MDAA)反應得到O-(2-羥基) 丙基-3-甲基-二丙烯基氨幾丁聚醣磺酸化物(O-HDACS),之後O-HDACS分別與4種N-(羧烷基)三苯基溴化鏻化合物(CATPP)進行醯氨化反應而得4種雙陽離子性四級鏻化幾丁聚醣衍生物(DCQPCS)。DCQPCS的特性以傅立葉轉換紅外光光譜儀(FT-IR)分析其分子結構,細胞相容性以L929纖維母細胞測試來評估其細胞成長活性與細胞毒性。抗氧化性則以DPPH游離基螯合能力來評估,另外,以金黃色葡萄球菌(Staphylococus aureus)測試最小殺菌濃度來評估抗菌活性。
In this study, the O-(2-hydroxy) propyl-3-methyl diallyl ammonium chitosan salt (O-HDACS) were applied to make reaction of chitosan and N-epoxy-2,3-propyl-N-methyl-N,N-diallylammonium methyl sulfate (MDAA). Afterwards, four double cationic quaternary phosphoniun chitosan derivatives (DCQPCS) were synthesized by O-HDACS and four (N-carboxyalkyl) triphenylphosphonium of compounds (CATPPs). These DCQPCS were characterized by FT-IR. Antioxidant activity were evaluated by DPPH (2,2-diphenyl-1-picryhydrazyl) radical scavenging assay. The cytocompatibility was tested by L929 fibroblast to evaluate proliferation and cytotoxicity. In addition, the antibacterial activity was tested against Staphylococus aureus by calculation of the minimum bactericidal concentrations (MBC).
