摘要: | 香茹(Glossogyne tenuifolia)為澎湖特有草藥,香茹茶是夏日清涼解熱及保肝飲料。許多研究發現香茹具有抗發炎、抗B型肝炎表面抗原表現及抑制多種癌細胞的增殖功效。本研究欲釐清香茹對先天性免疫的影響,因此使用巴豆醇-12-十四烷酸酯-13-乙酸酯(phorbol-12-myristate-13-acetate, PMA)將人類單核球細胞株,THP-1,分化成巨噬細胞系統進行研究。結果發現進行分化期間同時加入香茹甲醇萃取層(GT/MeOH),會增加巨噬細胞表面抗原CD11b及CD83分子的表現,顯示GT/MeOH可以增加巨噬細胞的分化。且分化時有GT/MeOH處理的巨噬細胞,即有ERK磷酸化現象,同時增加LPS誘發的第一型的干擾素(IFNα及IFNβ)的mRNA表現,由此可知GT/MeOH具有增加免疫的潛力。本研究證實香茹可以增加巨噬細胞分化,進而強化免疫力的作用,可以用於加強免疫力保健食品的開發。
Glossogyne tenuifolia, Hsiang-Ju, is a traditional antipyretic and hepatoprotective herb in Taiwan. A few researches reported that G. tenuifolia exhibited anti-inflammation, anti-HBV surface antigen expression and proliferation inhibition of cancer cells. This study investigates the effect of the methanol fraction of G. tenuifolia (GT/MeOH) on monocytic THP-1 differentiation into functional macrophage. Under phorbol 12-myristate 13-acetate (PMA) treatment, we found the surface markers, CD11b and CD83, of monocyte-derived macrophage (MDM) were increased with co-treatment of GT/MeOH. The MDM with GT/MeOH treatment during differentiation could produce much more IFNα and IFNβ mRNA by LPS stimulation. The phosphorylation of ERK by LPS stimulation was also increased on MDM with GT/MeOH cotreatment. In this study, we demonstrated that G. tenuifolia could enhance innate immunity via increasing macrophage differentiation from monocytes. The results in this study could provide strong evidence in the development of healthy food of G. tenuifolia on immunity promotion. |
