本計畫是以103 年度科技部計畫所開發的化學性鍵結成膜技術基礎下,製作軟性 的無機金屬氧化物為主的感測膜及以聚乙二胺樹枝狀聚合物為載體電極的電化學式農 藥感測器。內容分為兩部份,第一部份以低溫、簡單、便宜的自組裝法(SA)結合溶膠- 凝膠製程技術(SA-Sol-gel),以化學性固結法製作軟性的金屬氧化物無機感測薄膜,應 用於製備可撓式化學感測器的製作,將可開發新穎性的軟性材料,開發出各種化學感 測器。第二部份將利用新穎性的製膜技術, 層接式醯胺共價鍵結製膜技術 (Layer-by-layer and peptide reaction covalently anchored technique),來開發以聚乙二胺樹 枝狀聚合物為載體以鍵結乙醯膽鹼酯酶同時此聚乙二胺樹枝狀聚合物亦扮演著為H+接 受者之電化學電極應用於偵測水溶液中的農藥。 In this study, the soft inorganic sensing-film for flexible chemical sensors application and electrochemical pesticide sensors based on polyamidoamine dendrimer were fabricated using chemical covalently anchored technique. First, the flexible chemical sensors based on hydrolyzed three-dimensional sol-gel network inorganic materials were fabricated by combining sol-gel, nanoparticles and self-assembly technologies. The Au nanoparticles-graphene oxide-3-mercaptopropyltrimethoxysilane sol-gel (AuNPs/GO/MPTMOS sol-gel) and AuNPs/WO3/MPTMOS sol-gel films were fabricated. Second, a novel electrochemical biosensor for detecting methyl parathion and malathion was fabricated by covalently binding acetylcholinesterase (AChE), generation 5 amine-terminated PAMAM dendrimer (G5-NH2) and 3-mercaptopropionic acid (MPA) layer-by-layer on a gold electrode using N-(3-dimethylaminopropyl)-N′-ethylcarbodiimide hydrochloride (EDC) and N-hydroxysuccinimide (NHS) (EDC/NHS) as the peptide coupling reagent. The key step in the operation of this biosensor was the AChE-acetylcholine enzymatic reaction, which changes the local pH in the vicinity of the surface of an electrode.
