    Abstract: 本研究旨在探討山煙草癒傷組織誘導及增殖,試驗以不同濃度的2 ,4 -二氯苯氧乙酸[2,4-dichlorophenoxyacetic acid (2,4-D)]或1-萘乙酸 [naphthalene-1-acetic acid (NAA)] (0, 1.25, 2.5, 5 mg L^(-1))組合塞苯隆[1-phenyl-3-(1,2,3-thiadiazol-5-yl)-urea (TDZ)] (0, 0.1, 0.5, 1 mg L^(-1)),誘導山煙草根段、莖段及葉片培植體形成癒傷組織,莖段及葉片的誘導率均達100%,而根段的誘導率約20-100%,誘導形成的癒傷組織有淡綠色緊實及黃白色鬆散狀的型態。後者接續進行癒傷組織增殖的試驗,結果顯示不含植物生長調節劑或含2mg L^(-1) 2,4-D 的MS基本培養基有利於癒傷組織的增殖。試驗結果有助於山煙草癒傷組織誘導及增殖體系的建立,並進一步提供細胞及不定根懸浮培養系統建立之參考。
    In this study, the combinations of 2,4-dichlorophenoxyacetic acid (2,4-D) or naphthalene-1-acetic acid (NAA) (0, 1.25, 2.5, 5 mg L^(-1)) and 1-phenyl-3-(1,2,3-thiadiazol-5-yl)-urea (TDZ) (0, 0.1, 0.5, 1 mg L^(-1)) were used to test their effects on callus formation from explants of leaf, stem and root. Three weeks after culture, the explants of leaf and stem gave a higher callus formation rate than that of root. The developed calli showed two different morphogy, the pale green and compact calli or the yellowish white and friable calli. The calli from primary as secondary explants were used for further experiment of callus proliferation. Calli proliferation was better on MS basal medium containing 2 mg L^(-1) 2,4-D or auxin-free medium. The calli are suitable phytomaterials applied to the suspension culture of cells or adventitious roots, which can be used to isolate and purify bioactive steroidal alkaloids.
