試驗A以高血脂動物模式驗證桃仁及延胡索為主味藥之複方中藥之功效性。30隻雄性敘利亞倉鼠隨機分為3組:高血脂控制組、低劑量組(0.6 g/100 g bw 複方中藥)及高劑量組(1.2 g/100 g bw 複方中藥),三組皆餵飼一個月的高脂飼料,低劑量組於第零週開始餵飼中藥,高劑量組於第二週開始餵飼中藥,三組每週眼窩採血做血液採集並分析計算血液生化值。結果顯示,複方中藥對於高血脂倉鼠有降低膽固醇之功效。
試驗B以葛根及山藥為主味藥之複方中藥,研究對於由鏈佐黴素(streptozotocin, STZ) 所誘發之第二型糖尿病鼠高血糖之降血糖功效性。60隻雄性SD鼠隨機分為5組:控制組、糖尿病控制組、1倍劑量處理組(0.5 g/kg bw 複方中藥)、3倍劑量處理組(1.5 g/kg bw 複方中藥)及5倍劑量處理組(2.5 g/kg bw 複方中藥),除控制組外,五組皆以腹腔注射STZ之方式誘發糖尿病,在注射後一週確認糖尿病誘發成功(測量血糖值> 200 mg/dL);在第零週、第二週及第四週時以眼窩採集血液,在試驗第四週結束後做腹腔注射耐糖試驗。結果顯示,複方中藥對於高血糖SD鼠有降血糖趨勢,在耐糖試驗部分,結果顯示中藥處理組對於葡萄糖耐受性有改善之效果,並且有不傷害肝臟、保護腎臟功能及修復因糖尿病造成的胰臟功能缺失之效果。
Obesity and diabetes are both diseased characterized by hypertension, hyperglycemia and hyperlipidemia due to hard working, lack of exercise, stresses and bad eating habits at morden date and often leads to many other related complications. Effect of regular used medicine is usually harmful as compare to traditional Chinese herbal medicine. Therefore, it may be more moderate to use Chinese herbal medicine to reduce blood lipids and improve type I diabetes mellitus. The purpose of this study is to investigate two complex Chinese herbal medicines using hyperglycemia and hyperlipidemia animal models. Experiments have been done in two parts, one is using hyperglycemia animal model and the other is using hyperlipidemia animal model.
Experiment I investigates the effect of Complex Chinese herbal medicine (CCHM1) on the plasma lipid and cholesterol levels using hyperlipidemia hamsters induced by high-fat diet (HFD). Thirty hamsters fed with high-fat diet (HFD) were divided into three groups to test for the effect of CCHM1on the plasma lipid levels. The results showed that the hamsters fed with HFD resulted in high plasma total cholesterol (TC). The high-dose group is the most effective one. However, the CCHM1 did not show significant continuous effect after the medicine was withdrawn.
Experiments II investigates the effect of Complex Chinese herbal medicine 2 (CCHM2) on the plasma glucose and glucose tolerance test using streptozotocin-induced diabetic hamsters. Five-week-old male SD (Sprague-Dawley) rats (n=60) were used and 50 of them were treated by intraperitoneal injection of streptozotocin to induce diabetes. After injection, blood glucose was tested to confirm the successful induction of diabetes (> 200 mg/dL). Ten animals were untreated as control group. The results showed that the CCHM2 can improve glucose tolerance of the diabetic animals and is not harmful to liver, it also can help repairing the damage of pancreas caused by diabetes mellitus.
