    題名: 新月形黴菌毒素群之毒性體外評估系統建立與組合式黴菌毒素降解生物製劑開發-酵母菌與乳酸菌混合飼料添加劑在新月黴菌毒素處理之應用

    摘要: 酵母菌與部分乳酸菌對於黴菌毒素具有良好的吸附作用,能夠吸附和生物轉化黴菌毒素的特定乳酸菌更受到許多重視。若能進一步搭配酵母菌與乳酸菌,成為兼具益生菌功能的黴菌毒素處理添加劑,將對動物生產有更多方面的助益。本計畫之主要目標,希望結合具有刺激乳酸菌生長之特殊酵母菌與在動物腸道有效存活之乳酸菌,同時提供吸附與分解黴菌毒素的功能。希望在不改變飼糧特性及適口性的前提下,以較經濟之添加量來研發添加物,以解決新月黴菌毒素之問題。第一年計畫將由雞隻與猪隻消化道及糞便,篩選具有對抗新月形黴菌毒素Zearalenone (ZEA)與Deoxynivalenol (DON) 能力之乳酸菌株。篩選出之有效乳酸菌株與含有細菌素之酵母菌進行體外共同培養。第二年計畫將經過體外測試組合有效之含細菌素酵母菌及乳酸菌混合物,給飼仔猪驗證生長表現與腸道菌相狀態。具有潛力的組合,再加入特定黴菌毒素菌株接種後之仔猪飼糧,測定其生長表現,免疫反應之變化及腸道生理與微生物指標。第三年計畫結合瘤胃微生物來源解毒劑與混合菌添加劑,進行培養液測試及體外模擬消化試驗,以開發更全面的黴菌毒素解決方案。
    Yeast and some lactic acid bacteria (LAB) species have good adsorption ability to mycotoxin. However, LAB with adsorption and biodegradation activity has gathered great importance in recent years. Combining yeast and LAB to create a new mycotoxin treatment agent with probiotic activity may result in higher benefit on animal production. Aim of this study is to combine the yeast with LAB growth stimulation ability and the LAB with good proliferation performance in digestive tract, then making a treatment agent had both adsorption and biodegradation activity, offering a new product for Fusarium mycotoxin treatment base on the prerequisite of normal diet form and palatability. In the first year, LAB stains with decontaminating ability to Zearalenone (ZEA) and Deoxynivalenol (DON) will be isolated from animal faces and digestive tract content. The selected LAB strains will evaluate by co-culture with yeast contains bacteriocin (YB). In the second year, the effective mixtures of YB and LAB will apply on piglet feeding experiment to measure the growth performance and gut flora situation after in vitro evaluation. The mixtures have detoxification potential will add to piglet diet with Fusarium mycotoxin contamination in second stage feeding experiment. The growth performance, immunity response, physiology and microbiology parameter will collect to estimate the detoxification activity of microbial mixtures. In the last year, the detoxification agent from rumen ecosystem and microbial mixture in this study will be combined to precede co-culture and in vitro digestion test for creating better multi-function mycotoxin treatment agent.
