    摘要: 在近幾年的活體細胞分析中,細胞電阻抗分析已逐漸成為生物測量的有效方法之一。生物電阻抗特性可藉由細胞不同的電性特性的特徵響應,建立其活體細胞模型函數,可以提供比傳統生物病理組織分析更準確,更深入的細胞電性特性的信息。此外,為瞭解複雜的細胞反應,很多的細胞參數是需要在單一活體細胞才有辦法做量測的。因此在生物學與醫學的研究中,研究單一活體細胞特性,已經吸引不少研究人員的投入。本計畫擬利用單一細胞電阻抗量測建立一完整細胞電阻抗模型來分析細胞生理與病理上的資訊,以應用於癌症分析。在單一細胞捕獲部分,為避免在單一細胞捕獲時所造成之細胞損傷而造成建立細胞模型誤差,因此本計畫提出一種新型微結構設計進行機械式單一細胞捕獲捕獲,使用此種設計的細胞捕捉裝置,除了可以避免電力或熱力損傷細胞外還擁有高效率機械式單一細胞捕捉能力。而在細胞電阻抗模型部分,本計畫藉由改善量測電極靈敏度來減少量測上的誤差,在計畫中會進行不同電極材料與電極形狀分析,比較哪幾種電極較適合做為量測晶片使用、哪種電極可以得到最好的量測靈敏度結果,以達到最佳單一細胞電阻抗量測,再藉由模擬與實驗量測結果來建立一正確完整之單一細胞電阻抗模型。
    In recent years, the cell impedance analysis of living cells has gradually become one of effective methods of biological measurements. According to electric model function of living cells, the bioelectrical impedance analysis can provide more accurate, more in-depth information of living cell than conventional bio-histopathological analysis. In order to understand the complex cellular responses, the single living cell analysis is used. Therefore, a lot of group is already to study cells characteristics on single living cell level. This project proposes the electrical impedance measurement of single living cell to establish a complete cell electrical impedance model. The impedance model can be used to analyze the information on cell physiology and pathology and applied to the analysis of cancer. To establish the cell model error from single living cell capture, this study provides a new micro-structural design with mechanical type to single living cell capture. The new design device of single cell capture not only can avoid electricity or heat damaged to cells but also improve the percentage of cell capture. Moreover, the sensitivity ability of electrode is used to reduce measurement error in this study. To improve the sensitivity ability of electrode, the electrode shape design is used. Finally, the simulation and experiment measurement results is used to achieve the best single-cell electrical impedance measurement and create a correct, complete electrical impedance model of single living cell.
