    Abstract: 酵母菌與部分乳酸菌對於黴菌毒素具有良好的吸附作用,對黴菌毒素具有吸附力與生物轉化能力 的特定乳酸菌更受到許多重視。若能將可搭配酵母菌之可降解毒素乳酸菌,進行固定化提高作用能 力,將可成為兼具益生菌功能的黴菌毒素處理添加劑。本計畫之主要目標,希望利用具有刺激乳酸 菌生長之特殊酵母菌,結合來自動物腸道具有降解玉米赤黴烯酮(zearalenone, ZEA)之乳酸菌,以幾 丁聚醣將乳酸菌固定化,生產同時提供吸附與分解黴菌毒素功能的穩定型飼料添加劑。希望在不改 變飼糧特性及適口性的前提下,以較經濟之添加量來研發添加物,以解決動物飼糧中ZEA黴菌毒素 污染之問題。計畫將利用已由動物消化道及糞便所篩選具有對抗ZEA能力之乳酸菌株,與含有細菌 素之酵母菌進行體外共同培養,測試組合有效之含細菌素酵母菌及乳酸菌混合物,再將其中之乳酸 菌固定於幾丁聚醣,提升穩定性。完成之產品先經體外消化與代謝物細胞毒性測試,確認安全性後, 再添加加入含ZEA毒素之小鼠飼糧進行動物試驗,測定其生長、免疫反應與血液生化值之變化,並 檢測腸道生理與微生物指標。計畫將結合瘤胃微生物來源解毒劑與混合菌添加劑,共同進行培養液 測試及體外模擬消化試驗,以開發更全面的ZEA毒素解決方案。
    Yeast and some lactic acid bacteria (LAB) species have good adsorption ability to mycotoxin. However, LAB with adsorption and biodegradation activity to mycotoxin has gathered great importance in recent years. The immobilizated LAB with mycotoxin degradation ability and complementary to yeast could be applied to create a stable mycotoxin treatment agent with probiotic activity. Aim of this study is to combine the yeast with LAB growth stimulation ability and the gastric-intestinal source LAB with zearalenone (ZEA) biodegradation activity, then immobilizing LAB on chitosan to make a treatment agent had both adsorption and biodegradation activity. We attempt to offer a new product for ZEA treatment agent base on the prerequisite of normal diet form and palatability. The selected LAB strains will evaluate by co-culture with yeast contains bacteriocin (YB). The LAB in effective mixtures of YB and LAB will immobilize on chitosan to improve the stability and efficiency. The immobilized LAB and YB mixture then apply to in vitro digestion test and cell toxin examination. After confirming the safety of the product, the mixture will apply on mice feeding experiment to measure the growth performance, immunity response, clinic chemical parameters and microbiology parameter. The detoxification agent from rumen ecosystem and microbial mixture in this integrate project will be combined to precede co-culture and in vitro digestion test for creating better multi-function ZEA treatment agent.
