    Title: 利用幾丁聚醣3D多孔支架培養纖維母細胞之研究

    Abstract: 本研究是製作幾丁聚醣多孔支架並且觀察纖維母細胞在3D多孔材料上的貼附狀態及生長情況。此3D多孔材料是以幾丁聚醣為原料,NaOH為固化劑,利用不同的轉速製成各種不同孔徑的支架。3D幾丁聚醣支架經毒性測試,其實驗結果顯示3D幾丁聚醣支架不會對細胞造成傷害。3D幾丁聚醣支架經滅菌處理後植入纖維母細胞,以石臘包埋切片、染色的方法以及SEM觀察纖維母細胞的生長狀況。SEM觀察發現纖維母細胞確實能夠生長貼附在支架上。最後針對不同孔洞大小所得到的3D幾丁聚醣支架作細胞生長曲線分析,找出最適合纖維母細胞生長之3D幾丁聚醣支架與製作條件。

    This research studies the morphology and characterization of three-dimensional (3D) micro-porous structures produced from biodegradable chitosan for use as scaffolds for cells culture. The chitosan 3D micro-porous structures were produced by a simple liquid hardening method, which includes the processes of foaming by mechanical stirring without any chemical foaming agent added, and hardening by NaOH cross linking. The pore size and porosity were controlled with mechanical stirring strength. The scanning electronic microscopy (SEM) results showed that chitosan scaffold obtained by 4000 & 6000 rpm had the appropriate roughness for fibroblast attachment comparing with chitosan scaffold obtained by 2000 rpm.
