本研究以蝦殼為原料經酸鹼處理後製備成幾丁質,再經高溫熱鹼處理製備不同去乙醯化度之幾丁聚醣當作膠囊之殼材。將這些幾丁聚醣溶於2%的醋酸水溶液中形成幾丁聚醣膠體, 再以液中硬化法的方式包覆膠原蛋白的膠囊中,探討不同去乙醯化度幾丁聚醣之殼材的物性與成形膠囊的可行性以及膠囊中膠原蛋白的緩釋行為。幾丁聚醣經過黏度測試後發現去乙醯化度越高的幾丁聚醣膠體黏度會明顯降低。以膠原蛋白為芯材,使用去乙醯化度67.0%、81.2%、92.1%之幾丁聚醣作為殼材包覆,將包覆後的膠囊放入含有胰蛋白酶的PBS緩衝液中,以模擬人體中制放的行為,並比較這三種不同去乙醯化度幾丁聚醣為條件的制放曲線。
In this study, chitosan capsules containing collagen were prepared with various deacetylated chitosan wall materials by Orifice Method. The relation of deacetylated degree and viscosity of chitosan; the formation conditions of chitosan capsules and sustained release effect of collagen were investigated. The result indicated that the viscosity of chitosan was decreased with increasing the deacetylated degree of chitosan. The chitosan capsule can be obtained at the conditions of deacetylated degree 67.0%, 81.2% and 92.1 % of chitosan. The sustained release effect of collagen was improved over 100% by encapsulated with chitosan wall membrane.
